Is it possible to install the trunk tailgate speaker in a 2010 Jeep patriot that's not equipped with one? Also, is it possible for me to install a factory sub if not already equipped with one?
 
Welcome to the forum. I am sure someone here will have an answer for you. If it was not factory equipped with a sub then the wiring harness would not be installed, I don't believe but I am not an expert on this issue. also, the one that drops down from the tail gate on some models would have to have the harness installed at the factory as well. but no doubt any speaker system or sub can be installed. I was able to retrofit the Boston subwoofer on a 2011 Patriot. I will try to provide my experience so you can evaluate if is worth the effort or not.

First, note that the wiring is not compatible between the normal sound system and the Boston system that is the one having the factory sub in my 2011, so you will need to adapt it.

Then I gathered the sub enclosure and the plastic cover trim. In the 2011 non-sub there is a storage space in the trunk, and in the sub version the enclosure is placed there and a plastic cover with a grille is on top. That cover is plastic welded. With a heat gun or a pen iron solder (The solder used for electronics) can be removed. Is not difficult but is not a purchaseable part so need to be obtained from a junkyard unless you buy the whole rear right trim.

Also you will need a sub amp and a LOC (Low Output Converter) to connect the amp to the speaker line wires and then the sub to the enclosure. This is because the wiring of the original amp (That comes attached to the sub) is not compatible.

If done right, will look OEM, but is going to require a lot of work and money. I did it as I enjoy DIY projects, but in my opiniĂłn is better to get one sub enclosure that is designed to go under the seats.
